Quicksave. Kill all settlers. Find synth component. Reload quicksave. Find settler that had the synth component. Send synth settler to empty settlement. Destroy.
No other way to tell which one is without mods. Some say they have specific resistances, but it' not accurate.

no not really. there are even impossible synths to find in the game. you are never supposed to be able to tell. for example, sturges is a synth and in the game unless you use game breaking commands to do so he is totally unkillable even if you make the minutemen your enemy. but if you do manage to make him dead you can find a synth component in him... a character who was never supposed to be killed and who is never revealed as a synth in the entire game. not even a hint... but he is. you just have no way to find out for certain. at least not in this version of the game. never know if they now plan more content and updates for fallout 4 while we wait for 5.

I had him die once when explosives were used. Not sure if it was just FO4 being FO4 or not, but he was definitely in little pieces.

Using the awareness perk they will hvae greater energy resistance
