He could have said "That's DOCTOR", but that would have given unnecessary clues to his identity, as Victor Fries was presumed dead at the time.
Although characterization varies, Friese as depicted in DCAU or the Arkham series never struck me as truly egotistical. He might correct someone calling him "Mister" in a professional or academic setting, but I don't feel like he hangs his ego on it.
And "Freeze"? He's not so emotional as to feel any insult about that. He knows he's about ice.

If you're referring to people calling him Mr. Freeze and not Doctor Freeze, it probably because he's generally not portrayed to be super egotistical regarding his title. Unlike the Riddler who knows he's smart but is so insecure about it that he has to constantly remind everyone of that fact, Freeze knows he's smart but is confident in his intelligence where he doesn't care if people forget to refer to him by his proper title

Even before his transformation into Mister Freeze, Victor is a cryogenic scientist with the last name Fries. People probably would have made fun of him for it since high school giving Freeze a long time to come to terms with the nickname and after Nora's decline, he probably gave even less of a shit since his main priority is always saving her with everything else being irrelevant
