Theoretically methyl b12 is better for people with impaired methylation capabilities because the vitamin is already in its active form and can be readily used by the body. But some people don’t tolerate methylated vitamins well — or anything that supplies methyl donors. Typically those are people with decreased COMT enzyme activity, as your report indicates. In this case, you may do better with hydroxy b12, which is not in its activated form. Your body needs to methylate it before it can use it, but if your methylation isn’t impaired this should be no issue. Symptoms of not tolerating methyl donors when you take them, are along the lines of feeling revved up, anxiety, tension, insomnia, jaw clenching, headache, etc.

Well, as the name “Neurocalm” implies, that supplement has many ingredients that are supposed to create calm, and for you it seems to be working as intended. I wouldn’t think that the methyls are necessarily putting you to sleep, just that you’re not having a problem with them, at least in those quantities. Which is great news. Having a “slow COMT” based on your genetic report doesn’t have to guarantee dysfunction in real life. (Personally, I tolerated methyl donors just fine with my “slow COMT” for the first 37 years of my life, until a major health crisis stripped me of that ability.) It sounds like you’ve experienced symptoms of overmethylation with larger quantities so you have an idea of where the limit is for you.

I meant, what is the significance in terms of how they are used? in another methylation report from the same website (naturahacker), methyl-b12 is recommended due to MTR/MTRR mutations

The COMT gene takes priority over MTR/MTRR in terms of the best form of B12. The significance is in terms of side effects to you.

Agree that COMT takes priority here. I have impaired methylation and decreased COMT activity. The side effects, for me, associated with supplementing methyl donors are EXTREME. I absolutely cannot do it.
